Please note:
  • Melting temperature (Tm) prediction is for LNA and DNA/LNA oligonucleotides hybridized against perfect match DNA nucleotides in-solution only.
  • Each oligonucleotide must be separated by commas, or newline.
  • DNA monomers are lowercase a,g,c,t, and LNA monomers are uppercase A,G,C,T.
  • In this application LNA-C ('C') and 5-methyl-C ('mC') are equivalent.
  • Oligonucleotides should be longer than (or equal to) 8 and less than (or equal to) 20 monomers in length.
  • The Tm for DNA and LNA is calculated using a modified version of the thermodynamic model due to:
    John SantaLucia, Jr. (1998) Proc. Natl. Acad. Sci. 95 1460 - 1465 .
  • To see the effect of LNA on the Tm, try to enter the oligonucleotide as pure DNA, together with the mixed DNA/LNA oligonucleotide.
